Summary

Updates docs/WET_LAB_HANDOFF.md with three actionable wet-lab additions:

1. MDR strain panel table

Expands the MIC assay section to explicitly recommend testing against MDR clinical strains alongside ATCC reference standards:

  • MRSA USA300 — most prevalent clinical MRSA; any AMP hit is immediately publishable
  • E. coli ST131 (ATCC BAA-2469) — MDR ESBL Gram-negative, globally prevalent
  • K. pneumoniae ATCC BAA-1705 — carbapenem-resistant (KPC), WHO critical priority pathogen

Adds ~3× material cost but dramatically increases publication impact. Any candidate with MIC ≤ 8 μg/mL vs MRSA or KPC-KP qualifies as clinically significant.

2. C_AMIDATION_RECOMMENDED flag explanation

The presynth QC report (PR #36) now flags candidates whose C-terminus is neutral and charge pH 7.4 < 3.0. This entry in the flag table directs the synthesis team to request "C-terminal amide (CONH₂)" in the synthesis order form. Action costs nothing at SPPS ordering; adds ~+0.7 net charge and improves serum stability.

3. SEED-006 (Mastoparan-X) special notes section

Adds a dedicated section explaining:

  • Mechanism: calmodulin-binding / hydrophobic helix insertion (distinct from other 5 seeds)
  • Expected profile: Gram-positive bias; cooperative dose-response
  • Synthesis: no Cys/Met, amidation strongly recommended for LL C-terminus
  • Assay interpretation: novel mechanism confirmation if SEED-006 active while others fail

No code changes

Documentation only. All 872 tests pass on main.

Test plan

  • pytest — 872/872 passed (no code changes)
  • Document reviewed for scientific accuracy

…ANDOFF

Three additions:
1. MDR strain panel table: recommends MRSA USA300, E. coli ST131, KPC-KP
ATCC BAA-1705 for Wave 1 alongside ATCC reference strains. Any hit
against MDR strains is immediately publishable as clinically significant.

2. C_AMIDATION_RECOMMENDED flag explanation: directs synthesis team to
specify 'C-terminal amide (CONH₂)' in the synthesis order form.
Flagged for SEED-004/006 variants with neutral C-termini and charge < 3.0.

3. SEED-006 (Mastoparan-X) section: explains the wasp venom calmodulin-
binding mechanism, expected Gram-positive bias, hemolysis protocol,
synthesis notes (amidation strongly recommended), and interpretation
   guidance for novel mechanism confirmation.
